The place where you get the shuttle to the lighthouse is Drake's Beach.

 From my house in San Rafael, Limantour is a 1 hour drive, Drakes
takes 1.25 hours.
At Limantour, the walk from the parking lot to the water is 300 yards?
At Drakes, the walk from the parking lot to the water is 25 yards.
According to maps, the mouth of the estero is 2.5 miles from either
parking lot.

to launch at Drake's the wind is almost dead offshore, funnelling down
the canyon.
There are high wind-blocking bluffs on both sides, so the sailor must
get offshore a ways to get into steady wind.
But then, Tuba is a 10 minute highspeed zip downwind.
But then, the car is 2.5 miles upwind!
If you have a very good friend to drive the car around to Limantour,
that's another 45 minute drive!

At Limantour, the launch is side-off and the wind is steady. Upwind
the topography is dunes and rolling hills. You sail upwind first,
downwind to get home. If you don't make it all the way up to the
estero, you can still have a good time sailing right at the launch.

I have done it every possible way, and have settled on
the Limantour launch because it's a shorter drive and a safer way to do
it.
But an all downwinder with an extra driver is great way to do it.
Be sure to reward her well!
